摘要
采用简并引物反转录多聚酶链式反应(RT-PCR)和cDNA末端快速扩增(RACE)技术,从农业害虫铅灰齿爪鳃金龟中分离和克隆了一类非常保守的嗅觉受体蛋白基因(命名为Hp-Or83b)全序列。Hp-Or83b全长共1822 bp,5’-UTR为168 bp,3’-UTR为220 bp,开放阅读框全长1434 bp,编码478个氨基酸,预测分子量为54.24 kDa,等电点为7.46。通过同源性比较,发现Hp-Or83b氨基酸序列与已知的其它昆虫的Or83b具有较高的同源性。因此推断所获得的cDNA序列为农业害虫铅灰齿爪鳃金龟气味受体基因Or83b的cDNA序列。
An unusually highly conserved olfactory receptor gene(named by Hp-Or83b) was isolated and cloned from agricultural pest Holotrichia plumbea by reverse transcription-polymerase chain reaction(RT-PCR) and rapid amplification of cDNA ends(RACE) methods.The full-length sequence of Hp-Or83b was 1822 bp.Hp-Or83b contained 168 bp 5'-UTR,220 bp 3'-UTR and a 1434 bp open reading frame which coded 478 amino acids.The protein had molecular mass of 54.24 kDa and isoelectric point of 7.46.The sequence analysis indicated that the deduced amino acid sequences of the cDNA sequences shared high identity with Or83b orthologue receptor sequences from other previously reported insects.So it is inferred that the obtained sequences are cDNA sequences of odorant receptor gene Or83b from agricultural pest H.plumbea.
