摘要
研究了薄层扫描法及薄层-紫外分光光度法测定长春花中长春碱含量的两种方法,在硅胶F254-CMC-Na碱性板上(NaOH浓度为0.1mol/L),以氯仿-甲醇-石油醚(9:1:5)为展开剂,分离了长春花提取液中长春碱和长春新碱等14种成分。前法采用双波长簿层扫描,λs=280nm,λR=350nm,线性范围在2~10μg.后法采用薄层分离后,样品带用甲醇洗脱后,紫外分光光度法测定含量。后法在5~25μg/ml间符合比尔定律。两法回收率均在99%以上,RSD在2%以下,两法测定结果基本一致。前法较快速、灵敏、简便。后法不需特殊仪器设备。
This paper deals with two analytical metheds of vinblastine in Catharanthus roseus(L.)G.Don which are TLC scanning and TLC-UV.Fourteen components which were vinblastine, vincristine and so on in extract of Catharanthus roseus (L.)G.Don were separated on ailica gel F254-CMC plates with chloroform-methanol-petroleum ether(9:1:5)as mobile phase.Vinbtastine was determined by using dual wavelength TLC scanner at λs=280 nm,λR=350 nm.The calibration curve was linear in the range from 2 μg to 10 μg for the firot method and 5 μg/ml to 25 pg/ml for the second.The recoveries of both methods were bver 99% and their relative standard deviation was befow 2%.The former was fast, sensitive and simple.The latter didn't need special instruments and its precision wes good too.The extracting methed of vinblastine and the conditions for TLC developins were investigated.
