摘要
国产氨力农使培养乳鼠心肌细胞环腺苷酸含量和胞浆游离钙浓度显著升高且呈剂量依赖性;普萘洛尔不能阻止氨力农所致的环腺苷酸含量上升。
This study was to verify the effective mechanism of domestic amrinone (Am). After being treated with Am in different concentrations (50, 100, 300, 500 and 1000 μmol·Li(-1) n = 10 in each) the intracellular cyclic nucleotides (cAMP and cGMP) and cytoplasmic Ca(2+)([Ca(2+)]i) of cultured heart cells of neonatal rats were measured by radioimmunoaasay and fluorescent technology using Fura-2/am ,respectivly. As compared with the control group, there was significant concentrationdependent increase in both cAMP and [Ca(2+)]i (p<0.01)with significant linear correlation between them (r=0.86, p<0.01), while no significant change was found in cGMP. Propranolol (10 μmol· L(-1) )could not inhibit the increase of cAMP caused by Am. We conclude that domestic Am has the same mechanism of action as that of imported Am.
