摘要 人胚纤维母细胞在体外培养条件下,生长良好。群体倍增时间为1.93~2.36天;饱和密度3.5~4.4×10~4/cm^2培养面积;成集落效率0.7~3.3%;细胞周期为16.8小时,其中G_17.43小时,Ts5.2小时,TG_21.8小时,Tm2.37小时。HEF 在1640、199或加强Dulbecco Eagle 培养基添加10%小牛血清获良好生长。添加2%小牛血清加上10%羊水,对于HEF 生长支持作用与添加10%小牛血清相近,表明羊水中含有支持人类细胞体外生长的因子.HEF 具有容易培养,对遗传毒作用反应敏感等优点。可用于多种遗传毒理学研究·镍与砷化合物对HEF 显示了有明确剂量反应关系的细胞遗传学效应. A human embryo fibroblast(HEF)in vitro culture systembas been established as a model system for chemicalcarcinogenesis studies.The population doubling time of HEF is1.93-2.36 days,saturated density:3.5-4.4×10~4/cm^2,colonyefficiency:0.7-3.3%,the total cell cycle time:16.8 hours.TG(?),T_5,TG(?) and Tm are 7.43,5.2,1.8 and 2.37 hours res-Pectively.HEF is growing well in 199,1640 as well as Dulbe-cco Eagle reinforced medium supplemented with 10% calf bovineserum(CBS)or 2% CBS plus 10% human an(?)onic fluid(HAF),which indicated that there are some factors in HAF that cansupport the replication of HEF.After exposure to nickel andarsenic compounds the induction of chromosome abberation,sister chromatid exchange and micronucleus formation wereshown in HEF with a clear dose-effect relationship.As HEF iseasy to maintain and sensitive to different genotoxic effects,itpromises to be a useful system for genetic toxicology studies.
