摘要
本文进行了马洛替酯软胶丸和马洛替酯片在8例健康志愿者中的药代动力学及相对生物利用度研究,采用高效液相色谱法测定血清中具有生物活性的代谢物马洛替酯的单酯的浓度。结果发现,两种剂型在体内血药浓度-时间曲线均符合一室模型。马洛替酯软胶丸的主要药代动力学参数分别为:Tpeak=2.02±0.18h,Cmax=6.80±2.08mg/1.Tke=1.86±0.63h,AUC=39.06±11.52mg·h ̄(-1)·L ̄(-1)。其相对生物利用度为119.92%。
Pharmacokinetics and bioavailability of malotilate pill were studied with reverse-phase HPLC method. A single dose of pills and tablets (400 mg)was given to 8 healthy male volunteers in a randomized crossover study. It was found that the serum concentration-time curve of malotilate M- 1fitted to one-compartment model,and the main pharmacokinetic parameters were: Tpcak 2. 02 ±0. 18 h,Cmax 6. 08 ±2. 1)8 mg/1 ; T(ke) 1. 86 ±0. 63(h); AUC 39. 06 ±11. 52(mg ·h-1·L-1). Relative bioavailability of malotilate pill was 119. 92% .
