摘要
本实验选择日本大耳白兔,采用动脉内膜剥脱并喂养高胆固醇饲料的方法,建立兔髂动脉粥样硬化狭窄模型。结果显示:兔血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)降低,而高密度脂蛋白胆固醇(HDL-C)增高,血栓素 B_2(TXB_2)/6-酮-前列腺素 F_(1α)(TXB_2/6-k-PGF_(1α))增高。说明动脉粥样硬化的发生与内膜的损伤、脂代谢紊乱和 TXA_2/PGI_2失平衡有关。36只兔右髂动脉狭窄46处,主要是向心性局限性狭窄,程度为69%±25%,镜下可见内膜增厚,内有大量的脂质和泡沫细胞,胶原纤维和弹力纤维,以及平滑肌细胞。这种新的模型具有三个特点:(1)选用日本大耳白兔而不是新西兰家兔。(2)用球囊扩张导管代替 Forgaty 导管剥脱内膜。(3)胆固醇用量明显减少,每天只给0.5g。
44 Japanese white rabbits were divided into 2 groups:8 rabbits in control group (CG) were fed with common food.Right iliac arterial endothlium of 36 rab- bits in experimental group (EG) were denuded by balloon catheter and then were placed on 0.5g cholesterol diet mixied with 1.5g fat and 5g yolk every day for 10 weeks. Total cholesterol (TC),triglyeeride (TG),LDL-C and TXB_2/6-K-PGF_1α increased,and HDL-C decreased in EG.Iliac atherosclerotic stenosis (69±25%) in EG were found by angiography.Narrowing of arterial lumen, thickness of arterial endothlium in which there were lipid accumulation,proleferation of smooth mussle cells,colle- gen and elastic fibres were showed by light-and electron- microscopy. It was demonstrated that the model of iliac athroscle- rotic stenosis in rabbits is reliable and idea,and that the model has three characteristics (compared with others) in that (1) Japanease white rabbit was choose (New Jeland white rabbits).(2) balloon catheter was used (Forgarty catheter).(3) 0.5g cholesterol/day (2% cholesterol diet was given for each rabbit.It is also suggest that the for- mation of atherosclerosis be related to injury of endothli- urn,to abnormal TC,TG,HDL-C and LDL-C,and to imballance of TXA_2/PGI_2.
