摘要
为了进一步提高与稳定谷氨酸产生菌TH-89的产酸率,以谷氨酸棒杆菌TH-89为亲株,利用紫外线(UV)、硫酸二乙酯(DES)进行复合诱变和定向选育(耐高糖、耐高谷氨酸、以琥珀酸为唯一碳源),最终获得一突变菌株ZH-12.当初糖浓度为16%时,该菌摇瓶小试平均产酸率和平均转化率分别达9.85%和62.2%,而最高产酸率和转化率分别达10.9%和69%。
A brief introduction of the selection of Strain ZH-12 and its fermentative experiment ofglutamic acld is presented. A mutant of corynebacterium glutamicum ZH-12 is derived fromthe parental strain TH-89, by mutation with U V and DES.Under optimal culture condi-tions,when the enitial glucose is 16%the production of glutamic acid of the new mutant ZH-12 in flask is 9.85%,the conversion rate of glucose to glutamic acid is 62.2%;the maxiumproduction of glutamic acid reach 1 0.9%and the conversion rate reach 69%。
