人主动脉脂纹超微结构的进一步观察

摘要 为进一步探讨脂纹的超微结构,我们在以前工作基础上,用钌红染色做透射电镜检查另外11例主动脉的24对脂纹区及其附近正常区内膜。结果显示脂纹区某些内皮细胞(EC)糖萼明显变薄、有的EC退变甚至脱落、偶见细胞间隙扩张现象,进一步提示EC损伤和通透性增高在动脉粥样硬化(AS)中的作用。脂纹区有钙化小体,提示钙盐沉积可能开始于AS早期脂纹阶段。 In addition to our previous work (Chinese J Pathology 1985; 14:30), another series of specimens composed of 24 paired samples, i.e. one fatty streak with one adjacent normal tissue was taken from the aortic intima of 11 autopsied cases. Ten blocks sized 1mm^3 were cut from each sample. Ruthenium red stained sections were examined by transmission electron microscopy.The glycocalyx and glycoprotein in the pinocytotic vesicles and at the intercellular junctions of endothelial cells were ruthenium red positive. In fatty streaks, thinning of the glycocalyx and degenerative alterations of some endothelial cells could be seen. A few calcified bodies were occasionally found in the extracellular spacer and even less in the cytoplasm of intimal smooth muscle cells. A greater part of them was composed of concentric multilayered electron-dense material of both granular and needle shape which sometimes appeared as pseudoinclusion of an irregular shaped nucleus. Data support the possibility that enothelial injury is important in atherogenesis and calcium deposition may start at earlier stage of atherosclerosis.
