摘要
本文报告利用RP-HPLC与紫外检测器,以吡哌酸(PPA)为内标,快速测定人血清中依诺沙星(Enoxacin,ENX)浓度的方法。作者采用Ultyasphere-ODS分析柱,以0.1mol/L柠檬酸溶液(pH=2.5)-甲醇-乙腈(7:2:1)为流动相,波长选择265nm,以内标法定量。实践表明本方法具有灵敏、快速、准确等特点,适用于依诺沙星血药浓度的监测及临床药理学研究。
A rapid method for the reversed-phase high performance liquid chromatography using pipemedic acid as the internal standard was established.The method uses a 265 nm wavelength ultraviolet detector and reversed-phase ultrasphere-ODS(5μm,250 ×4.6mm)column,0.1m citric acid-methanol-acetonitril(7:2:1)as mobile phase.The detetor was operated at 0.2 AUFS.The retention times for pipemedic acid and Enoxacin were 4.4 9min and 5.74min respectively. Good linearity was obtained in the concentration range of 1-50μg/ml,with standard calibration curve y=12.0118x+0.1722 and the correlation coefficient r=0.9996.The detection limit was 0.5μg/ml.
