鱼腥藻HB1017株化能异养生长的研究 被引量:8

摘要 以葡萄糖和蔗糖为碳源,检测了六株(种)鱼腥藻的化能异养生产能力。其中鱼腥藻HB1017株化能异养生长较快,鱼腥藻HB0株化能异养生长缓慢,其余四种鱼腥藻不能进行化能异养生长。鱼腥藻HB1017株能利用果糖、葡萄糖、蔗糖为底物进行化能异养生长,但生长速率依次递减,差别显著。8磅湿热灭菌的果糖和蔗糖,与过滤灭菌的相比,只能维持低得多的化能异养生长速率。然而,8磅湿热灭菌的葡萄糖能维持比过滤法灭菌的高得多的化能异养生长速率,与过滤灭菌的果糖效果类似。添加外源精氨酸使异养生长速率明显提高。氮源以硝酸钠较好。25℃、30℃、35℃三组温度中,以35℃最好。适当的搅拌下生长速率比静置时高。光照生长的自养培养物转入黑暗中即可利用适当的碳源进行化能异养生长,无须经过“适应”过程。本实验中,3d最高平均生长速率为0.74,增代时间为0.9d。 Six species of Anabaena were initially examined fortheir ability to grow on glu- cose and sucrose chemoheterotrophically.Among these species,HB1017 grew fastand HB 0 very slow,While the other four species did not sutvive at all.Theeffects of fructose,glucose and sucrose,though all supported growth of HB1017,on growth were apparently different.The positive effect of the sugar substrateswas in the order of fructose,glucose and sucrose。It was found that autoclavedfructose and sucrose only supported a much lower heterotrophic growth rate thanthe filtrated ones,while autoclaved glucose sustained a higher growth rate asdid the filtrated fructose。The addition of exogenous arginine increased hete-rotrophic growth rate,and NaNO_3 was suitable nitrogen source for growth.Itappears that optimal growth was obtained if the culture was provided withappropriate stirring and temperature at 35℃。 The Photoautotrophic growth cultureof HB1017,when transfered to chemoheterotrophic condition,could quickly startto grow in it,without the“adaptation phase”.The average heterotrophic growthrate for 3 days was 0.74,and the generation time was 0.9 day.
