摘要
本文报道用新色原氯硝基苯基N-乙酰-β-D-氨基半乳糖苷(CNP-GAL)连续监测法测定尿GAL。此法不需尿样空白,有很高的精度,批内、批间的CV≤6.68%。尿GAL的最适pH4.4~5.0,最佳呈色反应pH4.6~5.0,在400秒观察期内吸光度变化与时间成正比例。ε=3.79×10~3L·mol^(-1)·cm^(-1),Km=0.37mol/L。TritonX-100可促进底物的溶解。葡萄糖、VitC、胆红素对本法无干扰。尿GAL正常参考范围:0.69~6.70U/gCr(n=158)。此方法适用于各种自动化分析仪,可用于临床肾病的诊断。
In this paper.we report to use a new chromogenic substrute 2-choro-4-nitrophenyl-N-acetyl-β-D-galactosaminide.and kinetic methed for determining the activity of N-aectyl-β-D-galaetosidase(EC 3.2.1.23.GAL) in urine by autoanalyzer.No sample blank is required. Very high precision is possible.with CV of≤ 6.68% both within-run and between-run. Chromogen CNP developed yellow color in pH 4.6-5.0.and the absorbance change during enzymatic hydrolyeations was measured automatically.The Km was 0.37 mmol/L.ε was 3.79×10 ̄3 L.mol ̄(-1).cm ̄(-1).Glucose.ascorbic acid. bilirubin do not interfere.The reference value of urinary GAL was 0.69-7.59 unite per gram of creatinine.The mothed is suitable for use with various autoanalyzers.and it should usefulfor diagnosis renal diseases.
