以HEV合成多肽为抗原制备抗-HEV免疫血清

Preparation of anti-HEV serum using the antigens of synthetic peptides

摘要 用对应于戊型肝炎病毒(hepatitisEvirus,HEV)开读框架ORF1、ORF2和ORF3部分氨基酸序列的三段多肽为抗原,以鼠伤寒沙门菌体为载体,经静脉免疫家兔和经腹腔免疫BALB/c小鼠,制备出抗-HEV免疫血清。两次加强免疫后,经本室研制的抗-HEV IgGELISA试剂盒检测,1:10稀释血清A值达1.50~2.00。用沙门菌和HEV多肽分别做中和试验,HEV多肽的中和抑制率大于50%,而沙门菌不能中和血清中的抗体。用此免疫血清与抗-HEVIgC阳性的病人血清做阻断试验,阻断率大于50%。上述结果证明家兔和小鼠产生的抗体为特异性抗-HEV抗体。 Three synthetic peptides corresponding to HEV genome OR1、ORF2 and ORF3,combined with SalmonelIa typhimurium were used as antigen to immunize rabbit and BALB / cmice. After two boost doses,the absorbance value of 1:10 diluted serum was 1.50-2.00 in thespecific ELISA test.HEV peptides could neutralize the antibodv at the rate of more than 50 %,whereas the Salmonella could not neutralize it.In addition to that.the immunized serum couldblock the conjugation of anti-HEV antibody from hepatitis E patients with specific HEV antigenat the rate of more than 50%These results show that the antibody produced by rabbit or mice isspecific anti-HEV antibodv.
