摘要
[目的]建立颞下颌关节骨关节病的动物模型,研究关节盘中弹力纤维、胶原纤维的分布及量的变化并探讨其与骨关节病的关系。[方法]20只新西兰白兔行左侧关节上腔穿刺,注射3%基质金属蛋白酶-10.1ml。于术后24h、1周、4周、8周、12周各处死4只,切取颞下颌关节标本。行Masson氏染色法和维多利亚蓝(VictoriaBlue)染色+范基森(VanGieson)复染法。在光镜下观察关节盘与双板区胶原纤维与弹力纤维的变化。[结果]注药后1周关节盘变薄,胶原纤维束变细,双板区弹力纤维散在断裂。第4周关节盘胶原纤维与弹力纤维排列稀疏、紊乱,部分断裂,双板区弹力纤维排列稀疏,大部分断裂。第8周关节盘出现穿孔。第12周可见胶原纤维和弹性纤维断裂、崩解。[结论]颞下颌关节骨关节病中胶原纤维和弹性纤维随病变发展而呈现逐渐加重的破坏性改变。
[ Objective] To establish animal medels to study the effects of fibres alterations on temporomandibular joint osteoarthrosis by investigating the changes in quantity and distribution of elastic fibres and collagen fibres of joint disk. [ Methods] 20 New Zealand rabbits were injected with 0.1 ml MMPs - 1 into the left joint cavity. Four rabbits were killed at 24 hours,1, 4, 8 and 12 weeks after the injection respectively. The joint were stained with VICTORIA BLUE and MASSON. Changes of collagen fibers and elastic fibers of joint disk and bioaminar zone were observed under microscope. [ Results] With VICTORIA BLUE dye the elastic fibers in bioaminar zone seemed arranged in disorder after the first week. Elastic fibers were sparse after the fourth week, a numbers of elastic fiber were broken ;with the MASSON stain collagen fibers in joint disk turned thin and disorderly arranged after the first week, collagen fibers were not running in various directions and were sparser in the 4th week as compared with that in the first week. the joint disks were perforated after the 8th week. With the MASSON and VICTORIA BLUE the collagen fibers and elastic fibers were ruptured after 12 week. [ Conclusion] With the development of TMJOA, the damage of elastic fibers and collagen fibers were aggravated gradually.
