念珠菌性外阴阴道炎患者阴道分泌物中白念珠菌分泌型天冬氨酸蛋白酶表达 被引量:1

Expression of Secretory Aspartyl Proteinase in Human Vulvovaginal Candidiasis

摘要 目的探讨白念珠菌分泌型天冬氨酸蛋白酶(SAP)家族在白念珠菌无症状性阴道寄居向症状性阴道炎发展过程中的表达。方法分别从10例无症状携带者、14例念珠菌性外阴阴道炎患者和10例复发性念珠菌性外阴阴道炎患者的阴道分泌物标本中直接提取RNA,逆转录成cDNA后进行RT-PCR。结果所有无症状携带者和患者均表现出对SAP2和SAP4-6的阳性扩增。10例无症状携带者中无一个标本表现出对SAP1和SAP3的阳性扩增,而念珠菌性外阴阴道炎患者和复发性念珠菌性外阴阴道炎患者阴道分泌物标本提取的RNA可见9个SAP基因的不同表达,并且均不同程度地表现出对SAP1和SAP3的阳性扩增。结论念珠菌性外阴阴道炎的发病可能与SAP基因的不同表达有关。 Objective To investigate the differential expression of secretory aspartyl proteinase in patients with vaginal C.albicans infection and in asymptomatic candidal carriers. Methods Secretory aspartyl proteinase expression was determined with revere transcription-PCR in vaginal specimens taken from 10 asymptomatic candldal carriers, 14 patients with vulvovaginal candidiasis (VVC) and 10 patients with recurrent VVC (RVVC). Results SAP2 and SAP4 to SAP6 subfamily were detected in both candidal carriers and patients with vaginal candidiasis. SAP1 and SAP3 transcripts were not observed in 10 asymptomatic candidal carriers. All 9 SAP genes were differently expressed in VVC and RVVC patients. SAP1 and SAP3 transcripts were frequently amplified in some patients. Conclusion it is shown that C. albicans infection is associated with differential expression of individual SAP genes, which may be involved in the pathogenesis of vaginal eandidiasis.
