摘要
用孔雀绿比色法可测出0.1μg磷,重现性好,变异系数为2.03%,回收率为94.4%~103.4%,显色稳定,操作简便。红细胞膜Ca ̄(2+)-ATP酶和Na ̄+、K ̄+-ATP酶经低渗皂素溶血后,在Ca ̄(2+)、EGTA或哇巴因的存在下,水解ATP释出的无机磷,用孔雀绿法可分别测出其酶活性。
Activities of Ca ̄(2+)-ATPase and Na ̄+,K ̄+ -ATPase of hemolysate were simultaneously deter-mined by a colorimetric method with malachite green dye. In the presence of Ca ̄(2+),EGTA or ouabain,inor-ganic phosphate released from ATP by hypotonic saponin-treated red cell lysate was used to estimate theactivities of Ca ̄(2+)-ATPase and Na ̄(2+),K ̄+ -ATPase. Themethod is simple,sensitive and stable for at least 4h.The effects of concentration of saponin and Ca ̄(2+),thestorage of hemolysate,and the activator and inhibitorof the enzymes on activity were discussed.
