富含组氨酸糖蛋白的纯化及促凝活性的初步研究 被引量:2

摘要 采用硫酸铵分段盐析,肝素-葡聚糖亲和层析和DEAE-交联葡聚糖A-50离子交换层析等方法从人血浆中分离纯化出富含组氨醚糖蛋白,经双向免疫扩散和SDS-PAGE鉴定,表明待鉴定样品为单一区带、分子量66Kd的富含组氨醚糖蛋白纯品。富含组氨醚糖蛋白通过同AT-Ⅲ竞争肝素结合而具有较强的促凝活性,其同肝素间的相互作用依赖于Ca2+的存在,EDTA,枸椽酸盐对此具有浓度依赖性的抑制作用。 n this paper. rnethods of ammonium sulfate fraction, heparin sepharose affinity andDEAE-Sephadex A-50 chromatography were used to isolate and purify histidine-rich gly-coprotein (HRG) from fresh human plasma. Analyzed by double immunodiffusion and SDS- PAGE , the purified sample produced a single precipitin arc when diffused against standardantiserum to human HRG. It showed a single band with an apparent molecular weight of66Kda, which was identified as HRG. The experimental studies revealed that HRG couldinhibit the anticoagulative activity of AT- Ⅲ and hence shorten the thrombin time. This ac-tivity depends on the presence of calcium ion and can be counteracted by calcium ion bindinganticoagulants such as EDTA and citrate. EDTA and citrate inhibit the interaction betweenHRG and heparin which have concentration dependent relationship.
