摘要
为探索空间条件对植物种子的诱变作用,对粳稻“农垦58”纯系单株种子经“8885”卫星搭载处理后在地面种植,筛选出已稳定遗传的大粒型突变系,对其基因组的突变位点进行了分子生物学分析,运用140个随机引物对“农垦58”原种及其大粒型突变系进行多态性研究,在扩增的2000多条DNA片段中,仅有5个片段显示了多态性,即所检测的2000个染色体位点上仅5个位点有差异,占0.25%,其中4个片段为重复序列,一个片段为2个拷贝序列。应用水稻窄叶青8号和京系17杂交F1代花药培养DH群体为作图群体,将该片段定位于第11号染色体上,位于分子标记CDO520和PTA818中间。
In order to investigate the mutation effects of outer space condition on plant
seeds,the cul-tivated rice“Nongken 58”seeds were carried into outer space by the
recoverable satellite“ 8885”in1988.The returned seeds were planted in the fields in Hainan
province. Large-grained mutant wasobserved in SP2 generation and selected. The mutant was
geneticly stable through six generations byself-cross. Molecular identification of the mutant has
been conducted by RAPD analysis, only five ofthe used 140 random 10-mer primers amplified
polymorphic DNA fragments between“Nongken 58”and its large-grained mutant.The
polymorphism degree is 0.25%. These polymorphic fragmentswere proved to be repeated
sequences except OPA18-3, which have only two copies in rice genome.RFLP mapping was
performed with a DH population from ZYQ8/JX17.Its location is on chromosome11 between the
markers CDO 520 and PTA 818. The sequence of OPA18-3 was also analysed.
