髓素碱性蛋白的提取及实验性过敏性脑脊髓炎动物模型的建立

Establishment of Experimental Allergic Encephalomyelitis Model by Using Myelin Basic Protein Extracted from Bovine Spinal Cord

摘要 我们从牛脊髓中提取髓素碱性蛋白(MBP)并进行电泳鉴定,证实与Sigma产品相似;然后用之作为免疫原免疫豚鼠,建立了实验性过敏性脑脊髓炎(EAE)动物模型,并对不同剂量MBP所致的EAE进行比较,选择出制备模型的合适剂量. Myelin basic protein (MBP) was extracted and purified from fresh white matter of boving spinal cord through a series of chemical procedures. According to the result of analytical SDS polyacrylanside gel electrophoresis, the quality of our MBP is similar to that of Sigma company. We established experimental allergic encephaiomyelitis (EAE) model using different doses of extracted MBP. We found that the clinical State of EAE was dose dependent. Our EAE model was very successful.
