嗜热脂肪芽孢杆菌质粒的研究

摘要 从广西各地的八个温泉的水样中分离到20株耐药的嗜热脂肪芽孢杆菌,用改良的BinghamSDS裂解法,从其中的15株菌中检测到质粒。再用琼脂糖凝胶电泳法及电子显微镜技术对耐氨苄青霉素(25μg·ml-1)的12号菌株的质地作了较深入的研究。发现该质粒为共价闭合坏状DNA分子(CCCDNA),共分子量为14.63×106,澳化乙锭(1μg·ml-1)和吖啶橙(20μg·ml-1)共同作用可将该质粒消除掉。该质粒的功能有待进一步研究。 Twenty a ntibiotic-resistant strains of Bacillus Stearothermophilus isoltatedfrom eight hot springs of Guangxi were exilmined for the pressence of plasmids by theBingham's modified method. Fifteen of 20 strains were found. to contain a singleplasmid. Through analysis by agarose-gel electrophoresis and electron microscopy, theampicillin-resistant Baeillus stearothermophihilus 12-was found to contain a plasmid ofmolecular weight 14.63 × 106(21 .95 kb). This plasmid is a covalently closed circular DNAmolecule (CCCDNA). It can be cured by ethidium bromide plus acridine orange. Its function isn't known. It's a cryptic plasmid.
