摘要
以人肝癌细胞株SMMC—7721为模型,用化疗药丝裂霉素(MMC),阿霉素(ADR),卡铂(CBBCA)和热休克处理后,观察其MDR1基因的表达情况。结果发现,SMMC—7721细胞经ADR处理后1周,MDR1基因表达即增加,MMC和CBBCA处理后2周,MDR1基因开始升高;而热休克处理后0.5hMDR1基因表达即见增高,在去除热休克因素后1h这种增高的MDR1基因表达开始降低。说明MDR1基因表达不但受化疗药的影响,也可以通过热休克进行调节人肝癌细胞中MDR1基因表达增加被认为是由于化疗药和热休克直接激活了MDR1基因启动子所致。所以MDR1基因也应是机体应激反应的主要基因之一。
The aim in the present study was to learn the relationship between chemotherapetic drugs and heat shock and the expression of MDRI gene in human hepatoma cell line SMMC-7721,which was treated by various chemotherapeutic drugs (MMC,ADR,CBBCA) and heat shock. The results showed that the ecpression of MDR1 gene was increased one to two weeks after treatment by ADR and MMC + CBBCA,respectively.However,the ecpression of MDR1 gene was increased 30 min after treatment by heat shock,and was edcreased 1h.after the suspension of heat shock. It is resonable to believe that the expression of MDR1 gene was influenced by not only the chemotherapeutic drugs, but also the heat shock.A possible mechanism of such phenomnon is that both drugs and heat shock can directly activate the promoters of MDR1 gene, and the latter could be considered as one of the main genes in stress rection.
