造血组织是辐射高敏感组织之一,胚胎动物的造血组织对辐射更为敏感。本文观察了持续性小剂量率γ射线照射妊娠中期母鼠,对出生子鼠股骨骨髓有核细胞数(BMC)、造血干细胞(CFU-S)数及其在股骨骨髓腔中分布状况的改变,和对造血组织基质细胞(CFU-F)的变化。本实验观察到正常小鼠股骨骨髓CFU-S分布,接近骨髓骨皮质的CFU-S浓度最高,约为41/105BMC,近骨髓中心静脉窦CFU-S浓度最低,大约是22/105BMC。60Coγ射线从妊娠13d到出生前持续性照射6d,分为15cGy/d剂量组和60cGy/d剂量组,出生子鼠成长到8周龄时,可见到股骨骨髓中CFU-S的分布紊乱,特别是在接近骨髓皮质部分的CFU-S数量明显下降,股骨骨髓中CFU-S总数也明显下降,15cGy/d组下降了20%,60cGy/d组下降了33%,骨髓基质中CFU-F数量在15cGy/d组下降12%,而60cGy/d组下降33%。统计学处理均有显著性差异。
The radial distribution of CFU-S in the femur of a normal 8 weeksold mouse is shown and corresponds closely to Lord and Hendry. The highest concentration (about 41/105 cells) was near to the bone surface and the lowest (about22/105 cells)was close to the central venous sinus which runs roughly along the longitudinal axis of the bone shaft. Continuous exposure to γ-rays for 6 d from 13dof gestation to birth had a smaller effect on the total femoral cellularity at 8 weeksof age, decreasing it by 12%-20%. The 15 cGy/d regimen caused a significant(20% ) CFU-S deficit (p<0.02) in the marginal zones. The 60 cGy/d scheduleincreased the deficit to a level (33%) and the change in the femoral CFU-S distribution after γ-ray irradiation is also shown. The 60 cGy/d schedule also reducedthe fibroblastoid, CFU-F POpulation, to 67% of control (P<0 .001). The 15 cGy/dreduced to 88% of control.
