摘要
报道了从人肝脏中纯化脯氨酰羟化酶β亚单位，５００ｇ肝脏经抽提、热处理、５５％～８５％硫酸铵分级分离后，再经ＣＭ－ＳｅｐｈａｄｅｘＣ＿５０和ＤＥＡＥ－Ｆｌａｓｔ－Ｆｌｏｗ柱层析得８０ｍｇ脯氨酰羟化酶β亚单位。ＳＤＳ－ＰＡＧＥ呈单一带，分子量为６０ｋＤａ。用纯的脯氨酰羟化酶β亚单位免疫新西兰大白兔获得效价为１：１６的抗血清。
AbstractPorlyl hydroxylase beta-stbunit has been isolated
from human liver.The preparation procedure in-volved heat
treatment,(NH_4)_2SO_4 precipitation,CM-Sephadex C_50 and DEAE Flast
Flow chromatorgraphy. The prolyl hydroxylase beta-subunit was
homogenous and has a molecular mass of 60 kDa by sodium
dode-cyl-sulphate-polyacrylamide gel electrophoresis. High titer
antiserum was preparaed by immunizing Newzealand rabbits with the
prolyl hydoxylase beta-subunit.
