摘要
应用Northern杂交技术发现遗传听源惊厥易感大鼠P77PMC在出生14天后脑内c-fosmR-NA水平迅速增高,21天达到最高水平,而后逐渐下降,此与大鼠惊厥易感性形成规律相类似;惊厥后脑内c-fosmRNA具有快速、大量及一过性表达特点;惊厥前后15分钟内给予安定(10mg/kg)能有效阻断c-fos基因表达。推测c-fos基因间接地参与了P77PMC大鼠惊厥易感性的形成。
e found that the basal c-fos mRNA level in the brain of P77 PMC rat(the audiogenic seizure-prone rat model)was rapidly reached the highest level on the 21st day after birth,then decreased to the same level with adult rat,which was correlated with the most susceptible period of audiogenic seizures.The character of c-fos mR NA expression in the brain of P77 PMC rat was rapid,large and transient after audiogenic seizure, Diazepam(10mg/kg,ip)injected 15min before or after seizure can completely block the c-fos gene expression.We suggest that c-fos gene is involved in the development and maturation of the central nervous system.There may be some close relationship between c-fos gene and the seizure susceptibility of P77PMC rats.
