摘要
本文以RPMI164O培养液为基础,探讨了补加因子及粘附胶原对大肠癌细胞体生长和贴壁的影响。结果表明:在基础培养液中添加10%的补充液可显著促进大肠癌细胞的生长,降低癌细胞对培养液血清的需求量;并表明,鼠尾胶原可明显促进癌细胞克隆形成率。对于组织贴壁培养,粘附胶原组成功率(66.7%)亦明显比无胶原被复组(41.7%)为高。
In order to improve the
sucessive rate of primary culture and sub-culture of large intestinal
tumors, supplements were added into RPMI 1640 culture me-dium and
collagen substrate was used to improve the anchorage condition. The
supple-ments included epithermal growth factor, pentagastrin,
transferrin, etc, and collagen sub-strate was prepared from rat
tails. The results showed that cell growth was improvedobviously in
PRMI 1640 contained 10%supplement media. though it was not
promotedwith increase of the concentration of supplements. This
supplement media could alsodecrease largely the requirement of cell
growth in vitro for serum, which may be help-ful to delav the growth
of fibroblast in primary culture. Though there were no relation-ships
between plate colonyforming rate and established cancer cell lines,
the existanceof adhesive collagen could increase the colony-forming
rate of primary culture and sub-culture. The survival rate of primary
explant culture in condition of collagen substance(66.7%)was higher
than that of the group without collagen coated(41.7%). It is
sugges-ted that optimization of culture condition be an important way
to Inhance the successfulrate of primary culture and subculture for
large bowel tumors.
