摘要
运用光敏生物素标记基因探针对23例新鲜肝癌和癌旁组织的c-mycmRNA和p53mRNA进行原位杂交(cDNA-mRNA)检测,发现在部分组织切片中存在内源件生物素样物质而产生假阳性,本实验采用一种封闭内源性生物素样物质的方法解决了假阳性问题,对这一方法的机理进行了讨论。
pairs of fresh tissues of hepatocellular carcinoma(HCC) and
corresponding non-tumorous liver(NT) had been studied by situ
hybridization(cDNA- mRNA)With photobiotin-labelled probes to
detect the expression of c- myc mRNA and p53 mRNA. It was
discovered that some cells in some sections showed endogenous
avidin-binding activity(EABA)which resulted in pseudo- signals. A
method of blocking EABA is introduced and its mechanism is discussed.
The results showed that it is a useful method.
