摘要 用胰蛋白酶和胶原酶消化法和组织贴块法培养猪肺动脉干内皮细胞和平滑肌细胞。方法简便易行,成功率高。培养的第2~3代细胞适用于缺氧实验和药物作用机制的研究。并可用于流式细胞术、同位素标记、单层细胞超微结构、免疫细胞化学和图像分析、原位杂交等的研究。因此,本方法培养的细胞可作为探讨缺氧引起肺血管构形重建及抗肺动脉高压药物疗效的一个体外研究模型。 Endothelial cells and smooth muscle cells of porcine pulmonary trunk were cultured by enzyme-dispersed and substrate-attached explant methods. These methods were simple,easy to operate, and efficient.Cultured pulmonary trunk wall cells at the 2nd to 3rd passage were applicable to investigations of hypoxic experiments and efficiency of drugs. These methods were applicable to flow cytometry, isotope label, monolayer cell ultrastructure,immunocytochemistry, image analysis and in situ hybridization as well. The cultured cells from these methods, therefore, can be used as an in vitro model for the investigations of structural remodeling of pulmonary vessels induced by hypoxia and efficiency of antipulmonary hypertension drugs.
