摘要
研究了创伤小鼠活化T细胞内游离钙浓度([Ca2+]i)、钙调素(CaM)、钙调素依赖的蛋白激酶(CaM-PK)活性的变化及其与T细胞功能之间的关系。结果显示,创伤后活化T细胞内[Ca2+]i降低,且这一变化同创伤小鼠T细胞白介素2(IL-2Rα)mRNA、IL-2受体α(IL-2Rα)mRNA水平降低,IL-2生成减少,IL-2Rα表达受抑,T淋巴细胞转化(TLT)降低密切相关。创伤后活化T细胞内CaM、CaM-PK活性明显降低。钙离子导入剂A_(23187)在体外可提高创伤小鼠活化T细胞内[Ca2+]i至正常对照水平,并明显提高其CaM、CaM-PK活性及IL-2mRNA、IL-2RαmRNA水平,但不能使之恢复正常。提示活化T细胞内[Ca2+]i、CaM、CaM-PK活性降低是导致创伤后T细胞功能受抑的重要原因。
The changes of free calcium level(Ca2+).calmodulin(CaM)and CaM-dependent protein kinase(CaM-PK)activity in the activated T-cells after trauma and their relationship with T-cell functions were studied in mice.It was found that Ca2+in activated T-cells was reduced after trauma,which was closely related to the reduction of interleukin 2(IL-2)mRNA and IL-2 receptor alpha mRNA levels,a decrease of IL-2 production,a suppression of IL-2 receptor alpha expression and a reduction of T-cell transformation in T-cells,CaM and CaM-PK activities in the activated T-cells were significantly decreased after trauma.Calcium cation ionophore A23187 could restore Ca2+level in activated T-cells of traumatized mice to the control level in vitro and it could also inncrease CaM and CaM-PK activities and IL-2 mRNA and IL-2 receptor alpha mRNA levels significantly though they were still under the normal levels.It is suggested that the reduced Ca2+and CaM and CaM-PK activities are the important factors to result in the suppression of T-cell functions after trauma.
