摘要
以AngraySK和自行设计的引物作半套式PCR,成功地从人外周血B淋巴细胞中扩增出Ig重链可变区基因。扩增产物的分子量为660bp左右。该文结果提示,在建立全套抗体基因文库时,半套式PCR法能较好解决Ig可变区基因多样性的扩增问题。
The variable region genes of the human immunoglobulin(Ig)heavy chains of peripheral Bcells were amplified with half-nested PCR using the primers designed by SK Angray and the authors themselves.The molecular weight of the PCR product was about 660 bp,The results suggest that when the library of the repertoires of antibody genes in to be constructed,half-nested PCR is useful to maximize the diversity of variable region genes of human Ig heavy chains.
