摘要
正常成人外周血细胞经二步法培养，可得到９０％以上纯度的红系细胞。并且红系细胞随培养时间延长，不断向成熟分化，红系细胞数目增多。以逆转录多聚酶链反应法检测该培养体系内珠蛋白链ｍＲＮＡ相对含量。结果表明：随着红系细胞成熟分化，细胞内β－ｍＲＮＡ表达升高，而γ－ｍＲＮＡ表达下降。表明二步法建立的红系细胞液体培养较好地模拟了红系细胞发育的生理状态，为体外研究珠蛋白基因转换提供了良好模型。
ononuclear cells separated from adult peripheralblood were cultured in a liquid system by two-stepmethod.During the period of culture,erythroid cellsproliferated with differentiation and maturation and,atthe end of culture(15 days after the begining of thesecond step ),more than 90% of pure erythroid cellswere obtained. Meanwhile,β-m RNAs ,γ-mRNAs andε-mRNAs were amplified by RT-PCR and quantitatedat day 5 and day 11,The results showed that theamount of β-mRNA increased while the γ-mRNA de-creased with ervthroid cells diferentiation,These re-sults indicated that this liquid culture systern well reca-pitulated the physiological status of erythropoiesis andcould be taken as a model for study the "switching"ofglobin gene expression in vitro.
