摘要
目的:研究创伤后肾脏组织中HIF1α表达的调控.探讨其在肾脏损伤机制中的作用.方法:从40cm高度自由落体打击大鼠脊肋角制作创伤模型,运用免疫组化方法进行HIF1α,TGFβ1染色.运用原位杂交方法检测HIF1αmRNA.结果:HIF1α,TGFβ1损伤后1,24h阳性细胞数较多,6,12h阳性细胞数较少.HIF1αmRNA各组表达均阳性但组间无差异.结论:HIF1α可能参与了肾脏损伤后缺氧的继发损害过程,并通过多个环节调控.对其进行深入研究可能为找到有效地治疗肾脏损伤的方法提供新的思路.
ATM: To investigate the expression of hypoxia-inducible Factor 1 Alpha in renal tissue posttrauma and to analyze its potential roles in the mechanism of renal injury. METHODS: The model used in the study was established by having the rats stricken by falling object from the height of 40 cm. Expressions of HIF-1α and TGF-β1 were measured with immunohistochemical staining and HIF-1α mRNA was measured with in situ hybridization. RESULTS: At 1 h and 24 h after renal injury, the number of HIF-1α and TGF-β1 positive cells was higher than those at 6 h and 12 h after renal injury. HIF-1α mRNA was positive in all the groups and no significant difference was found between the groups. CONCLUSION: HIF-1α may participate in the secondary hypoxic procedures after renal injury and be regulated by many factors. Further study of HIF-1α may provide some new therapeutic regimens for renal injury.
