EB病毒潜伏基因表达与系统性红斑狼疮的关系 被引量:3

Study on the Correlation Between the Expression of Epstein-Barr virus (EBV) Latent Genes and Systemic Lupus Erythematosus

摘要 目的检测EB病毒(EpsteinBarrvirus,EBV)潜伏基因在EBV阳性系统性红斑狼疮(systemiclupuserythematosus,SLE)患者外周血单一核细胞中的表达,探讨其临床意义。方法应用聚合酶链反应(PCR)Southern杂交技术检测44例SLE患者和43例正常对照者外周血单一核细胞中EBV特异性DNA片段BamHⅠW,筛选出EBV阳性标本,对EBV阳性标本进行RTPCR和Southern杂交,检测病毒潜伏基因(EBV核抗原基因EBNA2,潜伏膜蛋白基因LMP1和2A)的表达。结果44例SLE患者标本中有32例EBV阳性,正常对照组有3例EBV阳性,两组EBV阳性率比较差异有显著性(χ2=39.1779,P=0),SLE活动期和稳定期患者EBV阳性率比较差异无显著性(q=0.4025,P=0.7764)。32例EBV阳性SLE标本中有20例EBNA2mRNA阳性,1例LMP1mRNA阳性,但均未检测到LMP2AmRNA的表达。结论EBV感染参与部分SLE的发生发展,EBV潜伏基因EBNA2的表达可能诱发SLE患者的自身免疫反应。 Objective To detect the expression of EBV latent genes in peripheral blood monocytes of EBV-positive SLE patients and to understand its clinical significance.Methods The samples from 44 SLE patients and 43 matched normal controls were tested for EBV DNA BamHⅠ-W fragment by polymerase chain reaction(PCR)-Southern analysis .EBV nuclear antigen(EBNA)2, latent membrane protein(LMP)1 and 2A mRNA in EBV-positive samples of SLE patients were detected by RT-PCR and Southern blot.Results In peripheral blood monocytes of the 44 SLE patients,32 were EBV DNA positive,while 3 were found in control group.The EBV DNA in SLE patients showed higher positive rate than that of the normal controls (χ~2=39.1779,P=0),but no difference was found in the subgroups of SLE patients(q=0.4025,P=0.7764).To 32 EBV-positive SLE patients,EBNA2 mRNA was detected in 20 of the cases,LMP1 mRNA was detected in 1 of the cases,but no LMP2A mRNA in all cases.Conclusion EBV infection may participate in the pathogenesis of partial SLE patients,the expression of EBV latent gene EBNA2 may be related to the pathogenesis of the autoimmune reaction of SLE.
