摘要
目的探讨二氧化硒对体外培养的人卵巢癌细胞株COC1及其顺铂耐药细胞株COC1/DDP细胞增殖的影响。方法用MTT法检测体外培养的COC1、COC1/DDP细胞在单独SeO2作用下的增殖抑制及与DDP(顺铂)不同顺序联合作用下的增殖抑制,并以VRP(异搏定)为对照剂,对比SeO2与DDP不同顺序联合作用对COC1/DDP细胞的增殖抑制作用。结果①SeO2单独作用时对COC1、COC1/DDP细胞的增殖抑制作用随SeO2浓度升高而明显增强;②与逆转剂对照组VRP+间隔24h+DDP组相比,SeO2+DDP组及DDP+间隔24h+SeO2组抑制率明显高于逆转剂对照组(P<0.05),SeO2+间隔24h+DDP组抑制率与逆转剂对照组无明显差异(P>0.05)。结论在体外单独二氧化硒具有抑制人卵巢癌细胞株COC1、COC1/DDP细胞增殖的作用,二氧化硒与顺铂联合给药能明显增强人卵巢癌顺铂耐药细胞株COC1/DDP细胞对顺铂的敏感性。
Objective:To investigate possibility of overcoming the ci sp latin resistance in ovarian carcinoma cell line COC1/DDP induced by selenium di oxide(SeO_2).Methods:Growth inhibition of cisplatin-sensitive human ovarian ca ncer cell(COC1)and cisplatin-resistant cell(COC1/DDP)induced by either selenium dioxide combined with cisplatin or selenium dioxide alone were measured by me ans of MTT.Results:①The growth of COC1、COC1/DDP cell line induced by selenium dioxide were inhibited with dose-dependent pattern;②In all cases,the inhibitio n rate of COC1、COC1/DDP cell lines ranged from 68%(SeO_2+24 h+DDP group)、79%( DDP+24 h+SeO_2 group)to 80%(SeO_2+DDP grpup),and that of the latter tow group was similar(P>0.05).Conclusion:In vitro slelnium dioxide alone can inhibit growth of ovarian carcinoma(COC1、COCI/DDP)cell lines . Slelnium dioxide can in crease the sensitivity of COC1/DDP cell line to cisplatin.
