摘要
目的研究当归多糖(ASP)对免疫性结肠炎大鼠免疫功能的影响。方法建立大鼠免疫性结肠炎模型。ASP(250,500,1000mg/kg)灌肠用药21d后评价大鼠结肠粘膜损伤指数(CMDI),采用试剂盒、生物学方法及流式细胞术检测IL2、TNFα、转化生长因子β(TGFβ)活性,NO含量,外周血T淋巴亚群变化,及结肠组织髓过氧化物酶(MPO)活性。结果ASP灌肠给药降低模型组大鼠显著升高的CMDI及MPO活性,下调明显上升的IL2,TNFα活性及NO含量,显著上调降低的TGFβ活性,改善T淋巴细胞功能状态。ASP用药呈一定量效关系。结论ASP对免疫性结肠炎大鼠局部及全身免疫紊乱有一定调节改善作用,缓解结肠免疫损伤。
Objective: To investigate the immunoregulation of angelica sinensis polysaccharides on immunological function of rats with colitis.Methods:The immunological colitis model of rats was established.ASP(250,500,1 000 mg/kg)was used intracolonically for 3 weeks and the colon mucosa damage index(CMDI)was evaluated.The activity of myelopexoxidase(MPO),IL-2,TNF-α,TGF-β,the content of NO and T lymphocyte subsets in peripheral blood were measured.Results:ASP decreased the significant elevation of CMDI,the content of NO,and the activity of MPO,IL-2,TNF-α.In contrast,ASP markedly increased the sharply reduced activity of TGF-βin model group.ASP also improved the function of T lymphocyte in peripheral blood in model group.Conclusion:ASP shows immunoregulation on the immunological function abnormity of rats with colitis,reduce the immunological colon injury.
