摘要
为了观察冷冻速率和低温程度对组织细胞的损伤作用,将培养的大鼠主动脉内皮细胞(VEC)在不加任何低温保护剂的情况下,置于速率冷冻装置内,以1℃/min速率使培养液的温度分别降至0、-10、-20、-40℃或以10℃/min速率降至-40℃。在冻后1d和3d分别观察正常培养组和各冷冻组的细胞生长情况、培养液中乳酸脱氢酶(LDH)活性及VEC的因子Ⅷ相抗原(ⅧR.Ag)免疫荧光反应的情况。结果可见,冷冻后VEC的细胞数和生长率降低;LDH活性增加;各冷冻组的VECⅧR:Ag免疫荧光细胞数和阳性率降低。在同一冷冻速率下,上述这些指标的改变随冷冻温度的降低而加重;而在同一冷冻温度下,冷冻速率愈慢,损伤程度愈重。
In this study,the cultured vascular
endothelial cells( VEC )of rat’s aorta were frozen at different temperatures. The number
,growth rate of VEC ,lactic dehydroge-. nase(LDH )activity and the Factor Ⅷ related antigen(Ⅷ
R:Ag )in cultured medium were determined on the 1st day and the 3rd day after cooling.The
results showed that the number and growth rate of VEC decreased ,LDH activity increased ,the
number and positive rate of cells with immunofluorescence reduced after cooling ,and these
changes were closely related to the final temperature and the cooling rate. It indicated that
VEC were injured by low temperature even at 0℃in the absence of cryoprotector. The slower
the cooling rate and the lower the final temperature ,the more severe injuries were induced.
