摘要
目的:研究冷冻解冻、修复对金黄色葡萄球菌超微结构的影响,探讨冷冻损伤机理。方法:将正常未损伤、冷冻解冻、修复后金黄色葡萄球菌作负染、超薄切片,电子显微镜观察其超微结构的变化。结果:冷冻解冻后金黄色葡萄球菌的超微结构遭到破坏,胞内有空泡形成;金黄色葡萄球菌修复后,其超微结构基本恢复正常。结论:冷冻解冻可造成金黄色葡萄球菌超微结构的破坏,细胞膜损伤;金黄色葡萄球菌修复后,其超微结构可基本恢复正常;冰晶的形成、胞质浓缩产生的盐害是冷冻损伤的机理。
Objective:To study the influence of freeze-thaw and recovery on the ultra structure of the Staphylococcus aureus and the mechanism of the freeze-injury.Methods:The changes of the ultra structure were observed with the electron-microscope by scanning the minus dyed section and ultrathin section of normal,freeze-thaw and recovered Staphylococcus aureus. Results:The ultra structure of freeze-thaw Staphylococcus aureus was changed and the vacuole was formed in the cytoplasm;After recovery,the ultra structure of S. aureus was almost recovered.Conclusions:freeze-thaw can damage the ultra structure of Staphylococcus aureus and membrane was damaged;After recovery, the ultra structure of Staphylococcus aureus was almost normal;The ice-crystal and salt-harm which was formed by the concentration of the cytoplasm were the mechanisms of the freeze-injury.
