摘要
以限制性胃蛋白酶消化、分步盐析法从人胎盘中提取Ⅰ型和Ⅲ型胶原（ＣｏｌⅠ、ＣｏｌⅢ），用淋巴结内微量注射法免疫家兔，制备抗ＣｏｌⅠ和ＣｏｌⅢ的抗血清。抗血清经亲和层析免疫吸附后，ＥＬＩＳＡ和免疫组化检测证实所制得的抗ＣｏｌⅠ的ＣｏｌⅢ的抗体是单一特异的，它们的制备成功为组织学原位研究ＣｏｌⅠ和ＣｏｌⅢ的变化提供了可能。
As the interstitial collagen, type Ⅰand type Ⅲ collagen (Col Ⅰ and Col Ⅲ) are the main components of extracellular matrix. Because of the difficulties of collagen purification and antibody preparation, there are no anti-Col Ⅰ and Col Ⅲ antibodies available commercially so far. In this paper,Col Ⅰ and Col Ⅲ were extracted from human placenta by restricted pepsin digestion and a series of precipitation steps.Antisera were raised in rabbits and rendered specific for the immunizing antigen by immuno-absorption. These pnrified antisera were identified as monospecific ones by ELISA and immunohistochemical staining. There were no cross reaction between themselves and with the collagen of basement membrane.
