摘要
为了探讨神经元与其靶组织之间的相互作用及作用因素,建立了不同时期的纹状体外植块与14d大鼠胚胎中脑黑质神经元联合培养;14d大鼠胚胎中缝核外植块与黑质神经元联合培养。结果表明,生后2周、新生及14d大鼠胚胎纹状体外植块和14d大鼠胚胎中缝核外植块,对中脑黑质多巴胺神经元的形态发育有明显促进作用,其中包括长突起神经元数目和神经元突起长度的增加。上述3个时期的纹状体外植块,对中脑黑质多巴胺神经元的作用无显著性差异,而成年大鼠纹状体外植块则无作用。在形态学观察的基础上,还检测了中脑多巴胺神经元对外源性多巴胺的摄取力,14d大鼠胚胎纹状体和中缝核外植块能使中脑多巴胺神经元的3H-多巴胺的摄取力明显增高。以上结果初步表明,一定时期的纹状体和中缝核,对中脑黑质多巴胺神经元的发育有促进作用,并提示这种作用可能是通过释放神经营养因子而实现的。
n order to explore the interaction between netrons and their targets, two sets of experi-ments were conducted: (1) Striatal explants from different developmental stages of rat brainwerr cocultured with dissociated substantia nigra neurons from embryonic day 14 (E14) rat mes-encePhalon , (2) Raphe explant and dissociated substantia nigra neurons from E14 rat brain werecorvltured. It has been studied the effects of striatum and raphe nuclei on the development ofmesencephalic dopaminergic (DA) neurons. The results showed that striatal explants (from 14-day-old , newborn and E14 rat brain) and raphe explant (from E14 rat brain)promoted the de-velopment of mesencephalic DA neurons including the increases of two paramtters: (1) numberof nevrite-bearing neurons, and (2) neurite length. Among above three stages, the promotingeffect of striatal explants on the development of mesencephalic PA netrons had no significant dif-ference and striatal explant from adult brain was no effect on the development of mesencephalicDA neurons. On the basis of morphological study , we assayed [3H]: DA uptake in mesencephalicDA neurons. The result showed that the capability of mesencephalic DA neurons to take up[3H] DA was enhanced by striatal and raphe exhlants from E14 rat brain in coculture. These re-sults demonstrate that the development of mesencephalic DA neurons can be induced by striatumand raphe nuclei in certain stages and suggest that these effects may be resulted from the action ofneurotrophic factors released by explants.
