血管紧张素Ⅱ对高血压病NAD(P)H氧化酶P^(22phox)mRNA表达影响的初探

摘要 目的研究高血压病人血吞噬细胞NAD(P)H氧化酶P22phox mRNA的表达及其与血管紧张素(AT-Ⅱ)的相关性。方法研究对象分为原发性高血压组(EH)和正常组(N)。通过密度梯度离心法获取血中吞噬细胞,采用Rt-PCR技术检测吞噬细胞NAD(P)H氧化酶P22phox mRNA的表达量,放免法测定血浆AT-Ⅱ水平。结果高血压组血吞噬细胞P22phox mRNA较正常组高表达,血浆AT-Ⅱ水平的测定结果显示高血压组较正常组升高(P<0.01),两组人群血浆AT-Ⅱ含量与相应的吞噬细胞NAD(P)H氧化酶P22phox mRNA表达量呈显著正相关。结论高血压组血吞噬细胞NAD(P) H氧化酶P22phox mRNA较正常组有显著的高表达,推测吞噬细胞NAD(P)H氧化酶的活性可能受血浆AT-Ⅱ的调节。 Objective To study the expression of NAD(P) H oxidase subunits P22phoxmRNA in human blood phagocyte with hypertension, and the correlation with angiotensin Ⅱ. Methods Subjects were divided into two group, essential hypertension (EH) and normal (N). The phagocyte of blood were acquired by density gradient centrifugation analysis. P22phox mRNA were determined with reverse transcription polymerase chain reaction. The level of plasma angiontensin Ⅱ were detected by radioimmunoassay . Results The phagocyte NAD(P)H oxidase subunits P22phox mRNA expression were significantly increased in essential hypertension group in normal group. The level of angiotensin Ⅱ were significantly increased in hypertension group (P<0. 01). The results indicated that there were positive correlation in the phagocyte NAD(P)H oxidase subunits P22phoxmRNA expression with the level of angiotensin Ⅱ. Conclusion The expression of NAD(P)H oxidase subunits P22Phox mRNA were significantly increased in hypertension group. It is consult that the enzyme activity of NAD(P)H oxidase in phagocyte are regulated by angiotensin Ⅱ.
