摘要
本工作研究了人重组r-型干扰素包涵体的制备条件。用多次超声,差速离心等方法使包涵体与破碎的细胞结构和可溶性蛋白分离,使IFN-r占包涵体总蛋白的90%。通过用变性剂和透析等手段,使包涵体溶解,以利于IFN-r的进一步纯化。
In this work, the preparation process of inclusion bodies of HU-rIFN-r was researched.We adopted repeated sonication and differential centrifugation in order to isolate inclusion bodies from broken cell debris and soluble proteis. Because of the ease of isolation of the inclusion bodies HU -rIFW -r made up 90 % of the total cellular protein. By use of denaturants and dialysis the in clusion bodies were in solution.
