摘要
目的:应用动态比浊定量法研究头孢菌素类抗生素细菌内毒素。方法:通过对样本中定量添加标准内毒素的干扰预试验,将头孢菌素类抗生素制备成5种浓度系列,测得回收率,并筛选出最佳检测浓度,最后进行3个批号的试验。结果:内毒素的回收率分别为92.48%(头孢曲松钠舒巴坦钠),111.1%(头孢拉啶),99.39%(头孢他啶),73.31%(头孢曲松钠),111.9%(头孢呋辛),109.2%(头孢噻肟钠),133.3%(头孢哌酮钠舒巴坦钠),133.6(头孢硫脒)。头孢菌素类抗生素浓度小于等于1mg/mL的条件下,对动态比浊定量法无干扰作用,可用于有效的日常细菌内毒素检查。结论:动态比浊定量法为头孢菌素类抗生素的细菌内毒素的定量检测提供了一种全新的、高效的方法。
AIM:To study the dynamic turbidimetric limulus test used for endotoxin assay in cephalosporins for injection.METHODS:In the pre-interference experiments,the amount of endotoxin was added to 5 different concentrations of cephalosporins to get the recovery rates.And the optimum concentration was screened out.Under this condition,the interference experiments were conducted in three batches of cephalosporins(≤1 mg/mL).RESULTS:In the interference tests,the recovery rates of exdotoxin were 92.48%(ceftriaxone sodium and sulbactam sodium),111.1%(cephradimeinjection),99.39%(ceftazidimeinjection),73.31%(ceftriaxone),111.9%(cefuroxime),109.2%(cefotaxime),133.3%(cefoperazon sodium and sulbactam sodium for injection),133.6(cephathiamidiaum),respectively.It showed that solution of cephalosporins (≤1 mg/mL) didn′t interfer with limulus test.CONCLUSION:The dynamic turbidimetric limulus test could be conveniently used for endotoxin assay in cephalosporins for injection.
