摘要
目的:寻找渗透胁迫相关基因EST片断。方法:构建白花柽柳cDNA文库,对文库阳性克隆随机测序,并进行网上比对分析。结果:该cDNA文库克隆的重组率大于95%,插入片段主要集中在250~1000bp之间,并获得了如脯氨酸丰富蛋白基因,钙依赖蛋白激酶基因,丝氨酸苏氨酸激酶基因,过氧化氢酶基因,膜转运蛋白等基因。结论:渗透胁迫基因是多基因参与的系统调控过程,它们涉及了植物细胞壁合成、信号传递、基因调控、氧化物质清除、渗透调节物质的转运等方面。
Objective:To find the EST of Osmosis stress-associated genes. Method:a cDNA library was constructed from leaves of Tamarix albiflonum M. T. L iu .treated with osmosis stress. The picked positive clones of the library was randomly sequenced and analysed through BLASTX.Results:The rate of recombination was above 95% and the size of most inserts were 250~1000bp in the library and some genes were identified such as proline rich protein, calcium dependent protein kinase, serine/threonine-protein kinase, Catalase, integral membrane transporter protein etc.Conclusion:Osmosis stress-associated genes were multiple-gene that participated in systematic regulation process. The function of these genes included cell wall synthesis,signal component,regulatory protein and antioxidant enzyme.
