摘要
研究了长春新碱（ＶＣＲ）和甲基泼尼龙（ＭＰ）两药单用和联合应用对３种细胞系（Ｎａｌｍ－６、Ｄａｕ－ｄｉ和Ｋ５６２）的体外净化作用。利用多药效应分析方法定量分析了两药联合应用的效应关系。结果：两药以不同浓度比例联合应用对杀伤Ｎａｌｍ－６、Ｄａｕｄｉ细胞系克隆有协同作用，对前者杀伤作用最强。在达到同样杀伤效力时ＶＣＲ和ＭＰ两药联用剂量分别仅为单用时的８．７％～３２．３％和４．１％～２９．５％。两药联用杀伤Ｎａｌｍ－６和Ｄａｕｄｉ细胞系克隆近零时，ＣＦＵ－ＧＭ仍保留１７．５±３．５％。提示：两药联用对急性淋巴细胞白血病细胞系和淋巴瘤细胞系细胞净化效果显著，为ＡＬＬ患者自体骨髓移植（ＡＢＭＴ）用ＶＣＲ和ＭＰ体外骨髓净化提供了实验依据。
be purging effects of vincristine(VCR)or methyl-prednisolone(MP)alone and in combination on threeleukemia cell lines(Nalm-6, Duadi,K562 )were stud-ied. The effects of the drugs were analyzed with multi-ple drug-effect analysis method. The results showed thatthe purging effect of the combination of VCR and MPwith various molar ratios was synergistic for the Nalm-6and Daudi cell lines. Compared with VCR or MP alone,the dose of each drug in combination decreased 3.1～ 11. 43 and 3.4～24.96 folds respectively for obtainingthe same killing efficacy,The colonies of the Nalm-6and Daudi cells were declined to undetctale to undetectable level,while the CFU-GM still survived 17.5±3.5% in combi-nation of the two drugs. Our results suggest that thepurging effect of this combination in the autologous bonemarrow transplantation for acute lymphoblastic leukemiapatlents is promising.
