摘要
成人T细胞白血病(ATL)由人类Ⅰ型嗜T细胞病毒(HTLV-Ⅰ)引起,但其发病机理仍不清楚。我们应用逆转录-多聚酶链反应结合直接序列分析检测10例ATL患者的N-ras基因,结果在一例ATL发现N-ras61位点突变(CAA→CTA),说明HTLV-Ⅰ感染之后有多种因素参与ATL的发病。

Somaticmutation of N-ras is now considered to be a critical stepin human leukemogenesis. We tested N-ras gene pointmutation in 10 patients with ATL using reversetranscription-polymerase chain reaction (RT-PCR) toamplify N-ras cDNA containing codons 12 , 13 and 61 ,followed by direct sequencing. In one case, mutationwas found at codon 61 that resulted in activation of thisgene. The leukemic cells from the remaining ninepatients did not show N-ras point mutation. Theseresults suggested that multifactors rather thanmonofactor involved in leukemogenesis of ATL afterHTLV-Ⅰ infection.
