摘要
介绍胆固醇纯度标准物质的研制过程。用溴化一脱溴法纯化胆固醇,用高效液相色谱(HPLC)测胆固醇法和胆固醇氧化产物法检查纯化后胆固醇的均匀性和稳定性,用HPLC和差示扫描量热法及一系列辅助分析方法鉴定本品纯度和理化特性。结果证明本品均匀性良好,-20℃、氮气下至少可保存两年,纯度为(99.8±0.1)%。目前本品已被国家技术监督局批准为国家一级标准物质(GBW092033)。
he preparation of a cholesterol referenee material was described Commereial cholesterol was purified by treatment with broinine and dehalogenation with zinc.Homogeneity and stability of the purified cholesterol were exanuned by detennining cholesterol contents in different packages and moni-toring cholesterol oxidation products after periods of storage. Its purity was determined by high per-fonnance liquid chromatography and differential scanning calorimetry. A number of additional ana-lyses were also performed to characterize the material.It was homogeneous and stable for at least 2 years at-20℃under nitrogen.Its purity was(99.8 ± 0.1)%。 The material has been approved as the na-tional primary referenee material(GBW 09203a) by the NationaI Bureau of Technical Supervision of China.
