Simultaneous Determination of Cellular DNA/RNA and Distinction of G_0 from G_1 Phase with Acridine Orange Stain by Flow-Cytometry

摘要 应用吖啶橙(AO)检测细胞核酸含量,可同时获得DNA与RNA两项参数值.并根据两个参数确定细胞周期G0,G1,S,G2M的百分比值.AO与碘化丙啶(PI)两种方法测定药物对细胞周期的影响其结果基本相同.不同的是AO可以单独观察到G0期的变化,系用一个激发光源可获得两项参数的唯一染料. By using acridine orange (AO) for nucleic acid assay by flow cytometry, two parameters of celluar DNA and RNA were determined simultaneously. And in the light of the value of these two parameters,G0 phase could be identified and distinguished from G1, so that phase of G0,G1,S,G2M in cell cycle were examined in percentage.The results by AO assay were consistent with that by PI. AO is an useful and unique stain to observe G0 phase and to obtain two parameters of DNA/RNA with one excited light source in FCM.
