摘要
不同浓度的莪液对HeLa细胞有不同的毒性。选用合适浓度的莪对呼吸道合胞病毒(RSV)有直接灭活作用;对入侵细胞内的RSV可能有一定的抑制作用。提示用莪液向呼吸道喷雾治疗呼吸道合胞病毒感染,可能有效。建立了RSV感染的Balb/C小鼠的动物模型,并用莪静脉注射液进行了实验,治疗感染了RSV的Balb/C小鼠。在感染RSV的第5天和第8天,莪治疗组的鼠肺RSV分离率明显低于RSV感染组和葡萄糖对照组,P值分别<0.001和<0.0001,表明莪对感染RSV的动物有体内抑病毒作用。RSV感染组和葡萄糖对照组的鼠肺弹性及颜色改变明显不同于莪治疗组,P值<0.001及<0.003。对鼠肺组织用光学显微镜和电镜病理检查发现,在第5天,感染RSV小鼠的肺血管、毛细支气管区肺泡呈明显水肿和不同程度类症浸润,支气管上皮明显肿胀,可见不同程度的纤毛脱落和局灶性坏死,管腔内的巨噬细胞、多形核白细胞和纤维素形成栓子而致管腔阻塞,管腔内有明显的血小板聚集,肺泡间隔有嗜酸细胞浸润。葡萄糖对照组与RSV感染组的病理改变相同。在第5天,莪治疗组小鼠肺的毛细血管内皮细胞大部分正常,仅少数内皮损伤或形成囊泡,管腔内血小板聚集和淋巴细胞数量明显?
Ezhu is a kind of traditional Chinese medicine. In recent years it has been used to treat viral pneumonia in infants and young children with satisfactory therapeutic effects. The purpose of our experimental studies is to evaluate the effects of Ezhu on respiratory syncytial virus(RSV) which is the main etiologic agent of viral pneumonia in infants and young children.In vitro, the effects of Ezhu preparations on RSV in tissue culture were observed. The results showed that the drug could inactivate RSV directly and might inhibit virus replication after RSV entered the infected cells.In vitro studies, six-week-old Balb/C mice were divided randomly into four groups. The mice were exanguinated on the 5th and 8th days post-infection. The statistic difference was significant (P < 0.001 and < 0.0001) between the treated group and the control group. These experimeatal studies showed that intravenous preparations of Ezhu exerted good therapeutic effcts on RSV pneumonia without adverse reactions.
