番茄灰霉病菌的分子特征研究 被引量:1

摘要 选择来自上海的3株灰葡萄孢,扩增、测定其rDNA-ITS和RAPD种特异片段,并分析与之近缘的10种rDNA-ITS序列的差异性。结果显示:灰葡萄孢的ITS1、ITS2和RAPD种特异片段的长度和GC含量分别为147bp,42.18%/43.54%;160bp,46.88%;729bp,46.78%/46.36%。菌种间差异的范围:ITS1为0.014~0.102,ITS2为0.000~0.107。研究表明ITS序列和RAPD种特异片段均可作为灰葡萄孢鉴定的分子标志。 rDNA- ITS region and RAPD specific fragment of three strains of Bo trytis cinerea from Shanghai were amplified and sequenced. The ITS sequences of B. cinerea were aligned with ten sibling species. The results showed that length and GC contents of ITS1, ITS2 and RAPD specific fragment of B. cinerea were 147 bp, 42.18% /43.54% ; 160 bp, 46.88% ; 729 bp, 46.78 % /46.36% , respective ly. The intraspecific sequences of B. cinerea were conserved. The range of inter specific sequence diversity between B. cinerea and its sibling species such as B . porri, B. fabae and Sphaerotheca sclerotiorum were 0.014~ 0.102 (ITS1) and 0. 000~ 0.107 (ITS2). The results suggested that rDNA- ITS and RAPD specific frag ment were important molecular markers for the identification of B. cinerea. They could be used for early diagnosis tomato gray mould, if they were associated wi th other biological methods, such as morphology, Koch's Rule and relationship be tween pathogen and the host.
