摘要
目的建立测定栽培瓦布贝母西贝碱的高效液相-蒸发光散射检测方法。方法以Shim-packODSC18(150mm×4.6mm,5μm)为分析柱;以乙腈-水-二乙胺(60∶40∶0.3)为流动相,流速1ml·min-1;以蒸发光散射检测器(ELSD)检测,ELSD的气化温度为40℃,空气压力为1.7bar。结果西贝碱在0.1086~0.5430mg·ml-1之间线性关系良好,回归方程为lnC=0.7586lnA-10.183(r=0.9999,n=5);3、4、5年生栽培的瓦布贝母中西贝碱的含量分别为9.0%、8.1%、9.5%;平均回收率97.2%。结论所用方法快速、准确、简便,可直接测定栽培的瓦布贝母中西贝碱的含量。
OBJECTIVE To establish an HPLC-ELSD method to determine imperialine in F.wabuensis. METHODS HPLC was performed using Shim-pack ODS C_~18 (150 mm×4.6 mm,5 μm);Column,and acetonitrile-water-diethylamine(60∶40∶0.3) as the mobile phase,with flow rate of 1.0 ml·min^-1 .The ELSD was used as detector with evaporative temperature of 40℃ and air pressure of 1.7 bar.RESULTS Imperialine had a good linearity from 0.1086 mg·ml^-1 to 0.5430 mg·ml^-1 and the regression equation was lnC=0.7586lnA-~10.183 (r=0.9999,n=5). The contentsof imperialine in crude drugs cultivated 3,4,5 years were 9.0%,8.1% and 9.5%,respectively. The average recovery was 97.2%. CONCLUSION The method is rapid,accurate and simple.It could be used to determine imperialine directly.
