摘要
目的观察前列腺干细胞抗原(PSCA)在前列腺癌中的表达情况,探讨其与前列腺癌病理组织学分级及临床分期的关系。方法采用EnVisionTM两步免疫组织化学染色法,以抗人PSCA单克隆抗体检测正常前列腺、前列腺增生、前列腺癌及膀胱、肾脏等组织中PSCA的表达。结果PSCA在5例正常前列腺组织、10例前列腺增生组织、38例前列腺癌组织中有不同程度的阳性染色;膀胱、肾脏等正常组织的染色为阴性。95%(38/40例)的前列腺癌PSCA染色呈阳性、强阳性染色,50%~100%的肿瘤细胞着色,与前列腺增生及正常前列腺组织相比,其染色强度及范围的差异有显著性(P<0.01),但与前列腺癌的病理分级和临床分期不相关。结论前列腺干细胞抗原具有前列腺组织特异性,可用于前列腺癌免疫病理、放射免疫显像诊断及免疫治疗的研究。
Objective To examine prostate stem cell antigen(PSCA) protein expression in human prostate cancer specimens and to investigate the relationship between the PSCA expression and histologic grading and clinical staging of prostatic carcinoma. Methods A monoclonal antibody was raised against a Trx.6His-PSCA fusion protein and screened for their ability to recognize PSCA on the cell surface of human prostate cancer cells. 40 cases of prostate cancer, 10 benign prostate hyperplasia(BPH), 5 normal prostatic tissues and 5 non-prostatic tissues (including bladder,kidney,etc) were analysed for expression of PSCA, by anti-PSCA monoclonal antibody and EnVisionTM immunohistochemical staining. Results The antibody detected PSCA expression on the cell surface of normal and malignant prostate cells, but with regative results for other normal tissues. PSCA protein expression was identified in 38/40(95%) primary prostate cancers. There was no statistical difference in PSCA expression level between normal prostate tissues and BPH, but the PSCA expression level of prostate cancer was of significant statistical difference. There was no statistical difference between clinical staging and histologic grading of prostate cancer and the PSCA expression level. Conclusions The monoclonal antibody PSCA was organ specific and might be used for immunoimaging diagnosis and immunotherapy of prostatic cancer.
