趋化因子受体CXCR4在涎腺腺样囊性癌肺高转移细胞株中的表达 被引量:6

摘要 目的:观察趋化因子受体CXCR4在涎腺腺样囊性癌(adenoidcysticcarcinoma,ACC)肺高、低转移细胞株中的表达,探讨趋化因子受体CXCR4表达在ACC肺侵袭转移中的作用。方法:选取ACC肺高转移细胞株(ACCM)和低转移细胞株(ACC2)以及舌癌8113细胞株(Tca-8113),分别应用单克隆抗体免疫荧光标记流式细胞术和Western印迹检测CXCR4在上述3种细胞中的表达。结果:3种细胞表面均有CXCR4受体的表达,其中ACC肺高转移细胞株ACCM表达阳性率为(77.32±6.96)%,显著高于肺低转移细胞株ACC-2的(35.32±6.71)%和舌癌Tca-8113细胞株的(33.82±5.56)%,(P<0.05)。Western印迹结果证实3种细胞株均有特征性的蛋白质表达条带,其中ACCM的蛋白质表达条带最为显著。结论:趋化因子受体CXCR4在ACC肺高转移细胞株中高表达,提示CXCR4可能与ACC嗜肺转移的生物学特性有关。 Objective:To study the expression of chemokine receptor CXCR4 in adenoid cystic carcinoma(ACC) cells and evaluate the role of CXCR4 expression in pulmonary metastasis of salivary ACC. Methods: Fluorescence-activated cell sorter (FACS) and Western blot were applied for detection of CXCR4 expression in ACC-M, ACC-2 and Tca-8113 cell lines. Results: FACS and Western blot confirmed that there was protein expression of CXCR4 in the 3 cell lines, and that the positive cell rate in ACC-M cells([77.32±6.96]%) was significantly higher than that in ACC-2 cells([35.32±6.71]%) and Tca-8113 cells ([33.82±5.56]%,P<0.05). Conclusion: There is strong expression of CXCR4 in highly invasive ACC-M cells, indicating that CXCR4 might be related to lung metastasis of salivary ACC.
